RESUMO

The production and commercialization suitability of substance obtained by means of biotechnological processes depend on the separation techniques used for the purification of the desired compounds. Some features of the mixtures involved in these processes, such as low initial concentration, high purity level products desired, thermal sensitivity and the need of preserving the compounds properties, require a purification step that must be economical and biologically compatible with the chemicals involved. As a matter of fact, liquid-liquid extraction by aqueous two-phase systems is presented as an outstanding separation technique. The hydrodynamics and the mass transfer mechanism in the interior of the extractors are fundamental for the development of more efficient equipment. The more goai of this work was the study of a micro-column stirred by bell-shaped meshes for the extraction of a protein. The stirring system composed by the bell-shaped meshes promotes a better mixture of the fluids in the inner side of the column and, as a result, increase the mass transfer. The aqueous two-phase systems studied were formed by PEG (polyethyiene glycol) and salts (Potassium Phosphate monobasic and dibasic). It was done bath experiments, for finding the equilibrium concentrations and the better system for the extraction (PEG and salt concentrations and polymer molecular weight). The better condition obtained was used in the continuous extraction in the micro-column. As a conclusion, the extraction apparatus tested here had a much better yield of protein recovery than the observed in the batch system, what makes it suitable for use in biotechnologicai systems
