RESUMO

Candida species are opportunistic fungal pathogens that are often the causative agents of infections in immunocompromised patients. amphotericin B (AMPH), itraconazole (ITCZ), fluconazole (FLCZ) and 5-flucytosine (5-FC) are the principal drugs used on the treatment of candidemic patients. However, in some cases, the clinical responses of Candida are different from those obtained with the in vitro susceptibility tests. Moreover, the standard methods of monitoring the susceptibility of Candida to antifungal agents are unable to evaluate single cells, true hyphae or pseudohyphae forms. Thus, new techniques developed to measure the growth rate of true hyphae or pseudohyphae forms in vitro could contribute to better understand the dynamics of the formation of these invasive structures and the response of the organism to antifungal agents. Here we describe the application of an automated system (Bio-Cell Tracer® - BCT®) to examine the responses of Candida albicans to AMPH, FLCZ, 5-FC, as well as AMPH/FLCZ and AMPH/5-FC combinations, in simultaneous and distinct addiction times. The BCT® consists of a culture vessel, a microscope and a image analyzer with a computer program system that traces the growth of a hyphal tip automatically. The results obtained using the BCT® system were correspondent to those using the standard MIC (NCCLS A27) assay on the detection of susceptibility and resistance in clinical isolates and reference samples. The combinatory studies show an antagonistic pattern on the UN 219 strain, differently from the MIC studies. The possibility of tracing pseudohyphae structures allows a larger versatility and biological responses not found on the other tests. This work contributed with new information about an automated system for analysis of pseudohyphal structures, that may be used on the development of new strategies for a rapid and reliable assessment of antifungal susceptibility in Candida albicans or other infectious species
