Estudo comparativo dos efeitos do nimesulide e do acido acetilsalicilico sobre auto-hemostasia em ratos

RESUMO

The present paper has had as a purpose to evaluate the effects of an antiphlogistic non steroid, ( Nimesulide ) recently put on the pharmaceutic market about bleeding time (TS), coagulation (TC) and platelet counting in rats, comparing them to those provided by Acetylsalicylic Acid (AAS) whose interference in these hemostatic mechanisms has already been proved. For this purpose, 30 wistar, male rats ( 280 + ou - 10 g ) have been used, divided into 3 experimental groups with 10 animals each, which have received different treatment, orally taken : Group I : AAS ANTPOT. 1 5 mg/Kg unique dose; Group II : Nimesulide ANTIPOT. 2 3 mg/Kg in two doses on a 6 hour interval and Group III physiologic constituted serum in equívalent volume to the other groups (being in the control group). Blood platelet counting, the TS and and the TC were mesured before drugs administration (Zero time) in the following times: 2, 24, 48, 72 and 96 hours after medication. The results obtained has shown that AAS has increased the TS in all the times studied and TC as of 24 hours keeping alterated until 96 hours, after that however, there was no alteration in the numbers of platelet in none of the times, proving to be true trhe results of the early researches. On the other hand, with the Nimesulide althougt the tendency of TS increase has taken place it hasn t had any change in any of the period studied, nevertheless the TC has presented elevated in the times: 24, 48 and 72 hours. These results suggest that Nimesulide utilized in this dosage interferen with the hemostatic mechanisms, with lower intensity than AAS, in rats
