Efeitos do n-p (Azido-M- iodofeniletilamidoisobutil) Noradrenalina (Nain) "In Vivo e In Vitro" : um agonista de adrenoceptores beta

RESUMO

The selectivity of NAIN as a beta-adrenoceptor agonist was studied "in vivo" and "in vitro". The effects of the drug on arterial pressure and heart rate in conscious rats were compared with those obtained using isoproterenol. The pA2 value, obtained in isolated guinea-pig right atria for propranolol was compared to that obtained for metoprolol. The results from guinea-pig atrial preparations strongly suggest that NAIN activates both adrenoceptor subtypes since metoprolol was incapable of competitively blocking the chronotropic effect of NAIN (Schild Plot slope different from unity). These results are supported by the increase in heart rate observed in the rat right atrial preparation. In contrast, the experiments in conscious rats did not provide any significant evidence of receptor subtype activation, or was there a clear-cut dose-response relationship. The latter phenomenon may reflect, at least in part, the discrete bioavailability of the drug
