RESUMO

The aim of this study was to evaluate the effects of the treatment of spontaneouslyhypertensive rats (SHR) with lercanidipine on bone healing and bone density of the tibia, using histometric and TRAP expression analyses. Wistar and SHR rats were assigned to one of the following groups: Normotensive rats (NTR) (n=15); Untreated (n=15) and Lercanidipine-treated SHR (n=15). The latter group was treated daily with lercanidipine for 6 weeks. Two weeks after the beginning of drug administration, a critical-size surgical defect was created in the right tibia of all groups while the contralateral tibia remained without defect. The animals were sacrificed thirty days after the creation of a bone defect, corresponding to 45 days after the beginning of drug/placebo administration. The area of newly-formed cortical bone in the defect (bone healing BH), the medullar and cortical bone density (BD) of the tibia without defect and number of positive cells for tartrate-resistant acid phosphatese (TRAP) for both tibia were evaluated. There were no significant differences among groups for BH, medullar BD and number of TRAP+ cells in the defect (p>0.05). Significant higher cortical BDs and lower number of TRAP+ cells were observed for NTR and lercanidipine-treated SHR, when compared to untreated SHR (p<0.05). Treatment of SHR by lercanidipine during 6 weeks was able to increase the cortical BD and decrease the expression of TRAP+ cells.
