Efeito do tratamento com antioxidantes na qualidade de espermatozóides criopreservados de cães / Effect of antioxidants on post-thaw canine sperm quality

RESUMO

One of the main factors known to impair post thaw semen quality is oxidative stress (i.e., higher production of reactive oxygen species not compensated by improved antioxidant protection). The present experiment aimed to evaluate the effects of antioxidant treatment with glutathione (GSH) and vitamin E on the quality of dogs cryopreserved semen. Towards this aim, semen of twelve dogs was divided into mixes constituted of the ejaculate of three dogs with at least 70% motility. Each mix was then divided into 7 extender treatment groups as follows: Control, Vitamin E 1, 5, 10 mM, and GSH 1, 5 and 10 mM. O mix de sêmen foi então dividido em 7 alíquotas e diluídas em extensor a base de gema de ovo e ácido círtrico suplementadas com 0, 1, 5 e 10 mM de GSH e 1, 5 e 10 mM de vitamina E. After cooling, motility and vigor were assessed. Cryopreserved samples were thawed and evaluated for motility, vigor, percentage of sperm showing intact membrane (eosin/nigrosin) and acrosome (Simple stain), mitochondrial activity (diaminobenzidine) and susceptibility to oxidative stress (TBARS). Statistical analyses was performed using the SAS system for Windows. Samples treated with 1 mM of GSH showed higher percentage of intact membrane sperm when compared to the control (11.21 ± 2.84 and 6.21 ± 1.16, respectively; p<0.05). Treatment with 5 mM of GSH showed better results regarding mitochondrial activity. Effect of vitamina E treatment was observed only for the percentage of DAB III cels (i.e., cells with severe compromised mitochondrial activity), in which samples treated with 1 mM showed better results when compared to the control group (5.61 ± 0.7 and 8.62 ± 1.05, respectively; p<0.05). Results of the present study indicate that treatment with GSH and vitamin E to cryopreserved sperm samples of dogs improve post-thaw mitochondrial activity.
