Efeito do extrato acetato de etila de Cocos nucifera Linn (Palmae) sobre a resposta inflamatória e sobre a resposta imunológica in vivo. / Effect of ethyl acetate extract of Cocos nucifera Linn (Palmae) on the inflammatory response and the immune response in vivo.

RESUMO

The aim of this work was to investigate the effect of ethyl acetate extract of Cocos nucifera (EACN) through physiological parameters, topical inflammation induced by xylen and immune response in vivo. EACN was obtained from fiber coconut water and the phytochemical test indicated the presence of condensed tannins, flavonnons, flavonns, flavonnons, xantons and steroids. EACN was administrated to the Swiss mice orally in single dose, 0.2 mL, of 10, 30, 100 e 250 mg Kg-1 for five consecutive days to the toxicity protocols and topic inflammation. On toxicity experiments, it was observed the clinical and hematological parameters and macroscopic evaluation of the organs. The effect of EACN on topic inflammation was done in tests groups with different concentrations of EACN and positive control groups which received NaCl 0.9% or DMSO 5% or Dexamethasone (6 mg Kg-1, reference drug). All the animals received the flogistic agent (25 μL) in inner and outer parts of the ears two hours after the last treatment, except the negative control group. After 50 minutes from the xylen application, the animals were sacrificed, and a portion of each ear was took out and weighted. The difference between the weights of ears represents the effect induced by the treatments. To the protocols of immune response, mice were immunized with sheep red blood cells (SRBC) by followed treatments that were administered orally in single dose, 0.2 mL of EACN 10 and 100 mg Kg-1, NaCl 0.9%, vehicle, Levamisole (10 mg Kg-1) for 14 consecutives days or single dose of Ciclosporin (5 mg Kg-1). Antibodies titres were evaluated by hemmaglutination while cell immune response through delayed hipersensibility (DTH). EACN did not cause toxic effects, nor change hematological parameters neither organs relative weight. EACN did not cause inhibition of oedema induced by xylen, but in 250 mg Kg-1 presented pro-inflammatory effect (P<0.05). EACN inhibited primary and secondary antibodies production and did not inhibit DTH (P<0.05). EACN does not cause toxicity, nor anti-inflammatory activity neither altered relative weight of organs and it inhibited antibodies titres. In conclusion, EACN presented immunomodulattory effect on acquired immune response.
