Efeito do D-alfa-Tocoferol sobre a função renal de ratos hipertensos. / Effect of D-alpha-Tocopherol on the renal function of hypertensive rats.

RESUMO

Objectives: Evaluate whether or not D-alpha-Tocopherol has a protective effect on renal morphology and function in hypertensive rats. Methods: Wistar Kyoto rats (control) or SHR (hypertensive), male, 2 month old, were distributed in 4 experimental groups: C control treated with mineral oil; CT control treated with D-alpha-Tocopherol (CT); H hypertensive treated with mineral oil; and HT hypertensive treated with D-alpha-Tocopherol. CT e HT rats received D-alpha- Tocopherol (36mg/kg) in every other day, for 60 days. The arterial blood pressure was measured each 15 days. Kidney morphology and function were studied when the animals completed 4 months old. Results: In HT rats the blood pressure was smaller than in H rats but was increased in comparison to C rats. H rats presented lower values of GFR and RPF than C. Hypertensive rats treated with D-alpha- Tocopherol showed values of GFR and RPF similar to the control values. The urinary flow, the excreted amount of sodium and the excreted amount of potassium in H rats was smaller than in C rats but in HT these parameters were similar to C. In rats H the renal vascular resistance was increased in comparison to rats C. Hypertensive rats treated with D-alpha-Tocopherol showed values of renal vascular resistance similar to the control values. Morphological analysis showed that hypertensive rats presented glomerular hypertrophy and enlarged interlobular arterys wall. Conclusions: The D-alpha-Tocopherol treatment prevented glomerular and vascular hypertrophy. It also caused a decrease in arterial blood pressure and increased the excreted amount of sodium and potassium. The D-alpha-Tocopherol treatment improved renal and vascular function. These results suggest that D-alpha-Tocopherol has a beneficial effect on renal morphology and function of hypertensive rats.
