DivergÃncia genÃtica entre acessos de jaborandi (Pilocarpus microphyllus / Genetic divergence among jaborandi (Pilocarpus microphyllus) accessions

RESUMO

Jaborandi (Pilocarpus microphyllus) is a medicinal species that produces an alkaloid named pilocarpine, used for glaucoma and hair treatments, among others. The aim of this work is to study the presence of variability in the germoplasm bank of Embrapa AmazÃnia Oriental using RAPD and phenotypic markers. For RAPD analysis, it was sampled 93 individuals from 12 sample areas in the states of Parà and MaranhÃo. Pairwise similarities of Nei and Li were calculated for individuals and sample areas, then used to construct two dendograms and analyze intra- and intersample areas variance coefficients by analysis of molecular variance (AMOVA). The Shannon index was calculated for each sample area. For the phenotypic analyses, 152 individuals were sampled from 14 sample areas in the states of Parà and MaranhÃo. Leaves were sampled in two seasons, being analyzed the leaf dry weight, leaflets /leaf number, leaf length and width, length/width ratio and leaf area. It was obtained the multivariate variance analysis, using Wilks criterion. The canonical variables were obtained and used to calculate the Euclidean distances among sample areas. The average Euclidian distance was used to calculate the distances among individuals with standardized data. The analysis demonstrated that there is genetic differentiation between sample areas, and AMOVA demonstrated that 24,16% of genetic variation is maintained between sample areas and 75,84% is within, results comparable to others seen for herbaceous and alogamous species. The Shannon diversity index showed that Parà populations have greater diversity. The first three canonical variables explained 84,13% of total variation, and the six original variables were highly correlated to then and no variable was discarded. There was no correlation between distances obtained from phenotypic data and RAPD data.
