Desenvolvimento de biossensores para determinação de salicilato e glicose

RESUMO

The objective of this work was develop new techniques for biosensors construction. First of alI a biosensor for salicylate determination in drugs, after appropriate hydrolysis and dilution, was developed. A polypirrol film, obtained by electropolimerization dopped with hexacyanoferrate over which, a film of glutaraldehyde was added for enzyme salicylate hidroxylase ( EC 1.14.13.1 ), immobilization. Both films were supported on carbon glassy electrode surface, approximately 20mm. The biosensor showed a linear range of response for salicylate between 10 and 130 mmol L, at 150 mV vs SCE. The response time was 40 seconds. It was found a good correlation for the results obtained by the biosensor and standard colorimetric rnethod, in the assay of differents samples or antithermic drugs. In a second step, a biosensor for glucose determination, in samples of blood serum, was developed. For that, a modified carbon paste electrode was built, by using the glucose oxidase ( EC.1.1.3.4 )and ferrocene monocarboxylic acid, immobilized on silica gel-TiO2. The biosensor showed a linear range of response between 1.0x10 and 7.0x10 moI L, applying a potential of 340 mV vs SCE. The reponse time was 15 seconds. The determination of glucose in blood serum using the biosensor, showed a good correlation with the Merck "Kit" method. The immobilized enzyme showed an excellent stability, keeping its activity for at least six months.
