RESUMO

The Byrsonima intermedia A. Juss. is a medicinal plant from Cerrado, used mainly as febrifuge and astringent in diarrheas and dysentery, and present difficulties in relation to its sexual propagation. This objective of this work was to characterize the embryogenic callus obtained from leaf segments inoculated in MS culture medium supplemented with 1.0 mg L-1 2,4-D. After sixty days, the formed callus was replicated three times. For each sub cultivation, the number of globular embryos formed was evaluated. Samples of initial callus and those obtained from the sub cultivations were used for scanning electronic microscopy, starch and total phenols quantification, in addition to the determination of mitotic index and cell viability. The third sub cultivation showed the highest percentage of somatic embryos, approximately 70%. The cells changed the shape from elongated to isodiametric, characterizing meristematic cells with embryogenic capacity. Cells of the callus obtained in the third sub cultivation showed cellular organelles more developed, evidencing the embryogenic competence at cellular level. Callus sub cultivated showed a higher starch level at 60 days after incubation and the highest total phenolic concentration at 80 and 100 days cultivation. In the second sub cultivation, at 40 days of incubation, it was observed the highest cellular division index (18%) and at 50 days it was verified the highest percentage of viable cells (83%) with embryogenic characteristics.
