RESUMO
The cryopreservation of ovarian tissue has been the target of intense study with the purpose of assisting in the technology of assisted reproduction in animals and humans. For the success of cryopreservation, the composition of the freezing solution, especially the intracellular cryoprotectant agent used is of fundamental importance. Thus, this study aimed to define a solution for slow freezing of ovarian tissue goat. For this, two experiments were conducted. In experiment I, fragments of goat ovarian cortex were subjected to slow freezing in the presence of 12 different solutions containing 1.0 M ethylene glycol (EG), propanediol (PROH) or dimethyl sulfoxide (DMSO) as intracellular cryoprotectants, associated or not sucrose and / or fetal calf serum (FCS). In experiment II fragments previously frozen in solutions containing DMSO supplemented or not with sucrose and / or FCS were cultured in vitro for 48 hours. To assess follicle after thawing was used histology and viability test by fluorescent markers (ethidium homodimer-1 and calcein-AM) in experiments I and II, in addition, to ultrastructural analysis by transmission electron microscopy in experiment II regarding the statistical analysis, the effect of different cryoprotectant agents and interaction with supplements was used ANOVA followed by SNK test, while data concerning the viability were compared by chi-square. Values were considered significant when p <0.05. The first experiment showed that only solutions containing DMSO were able to maintain follicular morphology similar to that observed in fresh tissue. No significant differences were observed among the percentage of morphologically normal follicles in ovarian tissue cryopreserved before and after in vitro culture...
