RESUMO

This work aimed at assessing the chemical composition of the cell wall of isolates PT301 of Pisolithus tinctorius and H4111 of Pisolithus microcarpus, and see the effects of fungicides dimethomorph and pyroquilon in concentrations of 1, 5, 10, 15 and 30 ppm and chitosan in concentrations of 1, 2, 5 and 10 mg mL-1 on the micelial morphology. The wall of Pisolithus microcarpus H4111 presents itself consists of 46, 5 % of neutral sugars, 18,4 % of proteins, 26,1 % of chitin and 3.8 % of ash. The cell wall of Pisolithus tinctorius PT301 is made up of 48 % of neutral sugars, 17,7 % of proteins, 23,5 % of chitin and 4,5 % of ash. As to the mineral formation of the cell wall, it was found that the composition of nitrogen, phosphorus and potassium of the two isolates are similar, while the levels of sulfur and sodium are greater in isolate PT301. Within the levels of micronutrients, the fungus PT301 presented greater levels in zinc, iron and copper. The cell wall of Pisolithus has been rich in chitin throughout its length. The glicanes β-1,3 are found in greater quantities in the extremities of hyphae. The analysis of fatty acids showed that the composition of these compounds in the cell wall differs from that found in the mycelium, but in both situations, the most common fatty acids have been the 16:0, 18:1 w9c and mixing 18:0 A 18:2 w6,9c. Some fatty acids were detected only in mycelium and certain fatty acids were detected only on the walls. All three inhibitors used showed themselves capable of causing morphological alterations in hyphae of Pisolithus. They were checked swelling of hyphae, formation of vesicular structures at the end and also along the hyphae, and ripples and wrinkle of the mycelium. The fungicide dimethomorph also caused changes in connection clamps of the two strains.
