RESUMO

Inoculants containing N-fixer microorganisms generate huge economy for agricultural systems with nitrogen fertilization. In order to make inoculant products reach the market with the minimum attributes established in legislative determination, it is necessary to perform a good quality control. Therefore, this study aimed to compare methods of analysis of legume inoculants and to develop new methods that can be applied in the quality control of grasses inoculant products. Accordingly, 12 samples of legume inoculants were analyzed ¿ seven liquid and five peaty ¿ along with one sample of grass inoculant. The serial dilution methods were compared to those determined by the legislature, using different types of diluent, tube agitation, and plate inoculation methods. It was also evaluated the survival rate of microorganisms inoculated in soy and corn seeds up to 96h after inoculation, and the use of glass spheres as an alternative for soy seeds in survival tests. Using a vortex for 10s showed the most gainful results for use in serial dilution methods. There was no significant variation between the diluents used, so the distilled water was considered the most convenient alternative. The drop plate technique was the most efficient due to the lack of difference between inoculation methods. The glass spheres and soy seeds did not show actual difference in number of viable cells recovered. There was a reduction in number of bacteria inoculated in soy and corn seeds after 6h of storage. As a result, the methods for serial dilution, plate inoculation, and evaluation of inoculated seeds can be optimized for quality control of inoculants, allowing more convenient laboratory procedures for the operators.
