Caracterização molecular de cepas de Staphylococcus aureus isoladas no Hospital Municipal de Ipatinga-MG

RESUMO

Infections caused by methicillin-resistant Staphylococcus aureus can have serious consequences for patients of health services. Several control measures are implemented to contain the spread of these microorganisms, but not always sufficient. This work was identifying strains sensitive of Staphylococcus aureus (MSSA) and methicillin-resistant Staphylococcus aureus (MRSA) belonging to the database of the Clinical Laboratory of the Municipality of Ipatinga / MG isolated from clinical specimens from patients colonized with S. aureus hospitalized from april 2008 to april 2009. During one year was identified 439 isolates and these set, 50 (11.4%) showed morphological and phenotype characteristic of S. aureus and 6 (12%) isolates were resistant to at least one antibiotic while no strain was resistant to vancomycin. To analyze the population structure of S.aureus we using PCRDenaturing gradient gel electrophoresis (DGGE) approach. Initially the genomic DNA was been extracted and amplicons specific to V3-V5 regions of the bacterial 16S DNAr were obtained and analyzed by DGGE. Our results suggest a high genetic variation in S. aureus isolates. The MRSA strains were further characterized by testing for various multi drugs genes using qRT-PCR. Our results showed a variation on profile of expression between mecA-1, mecR1-2, mecl3, norA4 and norA5 genes in MRSA strains isolated, suggesting that the profile of expression can be influenced by the genotype variation observed among these isolates. We concluded that various clones of S. aureus was been circulating in the patients from Ipatinga. According to our results, future efforts should be directed toward optimizing the use of PCR-DGGE as a toll to rapid identified S. aureus.
