RESUMO

The dengue virus is more important disease transmited by mosquitoes in terms of morbidity and mortality. The Aedes aegypti is the unique vector of the epidemiologic importance for dengue virus transmission in the America. The symptoms of dengue are characterized by acute febrile, accomplished of headache, retro-orbital pain and muscle and bone or joint pains about others symptoms would can developed benign disease or dengue hemorrhagic fever and dengue shock syndrome (DHF/DSS).Rondonia State is located in north region of Brazil and considerate appropriate for introduction of news serotype and the outbreaks of virus because had the borderline extent and cutting by road BR 364 which linked the Acre State the others parts of Brazil. In the road are intensive traffic of trucks, bus and cars which make easy the transport of infected peoples. The epidemiological survey is important for detect the new serotypes of dengue virus introduced by infected peoples which cross the State. The methods for characterization and molecular serotyping of dengue virus are helpful for detection of the introduction of new serotypes and prevention of outbreaks. The molecular studies helped in the establishment of the correlation of disease and dengue virus strain. In this work will describe how types of dengue virus circulate in Rondonia State. Eighty four samples of blood of the patients with dengue virus suspect were collected between January/2004 and February/2005. The serums were inoculated in C6/36 tissue cells by seven days. After, the fluid is colleted and the RNA extracted by TRIzol method and maked the RT-PCR with FG1 and FG2 universal Flavivirus primers.The serotypes of dengue virus were characterized by hemi-nested-PCR utility the specific primers nden-1, nden-2, nden-3 and nden-4. The other primer pair was utility for RSS-PCR. The nucleotide sequence was obtained in the MegaBACE 1MB. The results were: forty four samples with dengue virus type 3. The amplification by RSS-PCR generated fragments corresponding at the group C. Five aleatoric samples had sequencing and compared, by clustal W and BLAST sofwares, with others sequences obtained in GeneBank corroborating with others results in the specialized literature. Therefore, the dengue virus circulating in Rondonia State is type 3, with origin in the America.
