Caracterização da atividade enzimatica da triparedoxina peroxidase em Trypanosoma cruzi

RESUMO

The objective of this work was to charactetize and compare the tryparedoxin peroxidase (TXNp) enzymatic activity in two strains of T. cruzi that have different resistances to oxidative stress. TXNp identification was made in total ceIlular extracts ITom epimastigote of Y and Tulahuen 2 strain. In a broad sense, Tulauen 2 strain has a higher concentration of this enzyme when compared to the Y strain. Differences in TXNp concentration into a same strain with different geographic origins were also observed. TXNp present in total ceIlular extracts ITom T. cruzi epimastigote has an antioxidant function, protecting glutamine synthetase (GS) against reactive oxygen species (ROS) produced by the DTTlFe3+/02 oxidant system. lt appears not to be involved with cellular proliferation, since a correlation could not be stablished using Westem Blot analysis, between the different growth days and the amount of this protein. Tryparedoxin (TXN) was also found in the same extracts and variation in its concentration were observed when strains and different growth days were compared. Also in this case, Tulahuen 2 ceIls had a higher concentration of TXN. The higher resistance observed in the Tulahuen 2 strain against ROS generated by DTTlFe3+/02 system can be explained by the higher concentration of TXN and TXNp found. This would led to a hydroperoxide metabolism system more effective, when compared to the Y strain. T. cruzi sheddings of both forms (epimastigote and trypomastigote) showed liberation ofTXNp to the extraceIlular environrnent
