RESUMO

The cryopreservated semen at artificial insemination programs is a value tool for genetic and ovine breed conservation programs. To facilitate the disponibility of seminal extenders and reduce the costs, it was developed a vegetal extender based on powder coconut water (PCW- 102), that shown satisfactory results in chilled ram semen. However, there is a lack of studies using this extender in ram semen cryopreservation. The aim of this study were to evaluate the efficiency of ram semen cryopreserved on PCW-102c and TRIS on sperm motility and sperm head morphometry using computer assited semen analysis (CASA). Twelve semen collections were taken from four Santa Ines rams. Semen samples of each ejaculate was divided into two aliquots and extended on PCW-102c and TRIS and cryopreserved. The semen was evaluated at the moment of thawing (T0), after one hour (T1) and two hours (T2) of incubation, for percentage of spermatozoa morphologically normal and alive cells (by eosin-nigrosin dyeing technique), evaluation of acrosomal integrity and cellular viability (by triple stain technique) and motility evaluation at CASA system. Fresh and frozen-thawed semen were smeared and stained in Bengal Rose dye and analyzed using the morphometric module of CASA system. At least 150 properly digitized sperm head per slide were analyzed for length, width, area and perimeter. It werent observed statistical differences between semen cryopreserved on PCW- 102c and TRIS extenders for the parameters adopted in each stain technique. There are significant differences at the moment of thawing between PCW-102c and TRIS extenders for total motility, progressive motility and head lateral amplitude, with superior values for TRIS extender, but both extenders was equivalent for qualitative (straightness and linearity) and quantitative parameters (curvilinear velocity and average velocity), and frequency of track crossing. After two hours of incubation, both extenders were significantly different for motility parameters evaluated, except for linearity. The PCW-102c extender shows more individual variability between rams. The morphometric parameters for fresh semen were statistically superior to those of frozen-thawed semen, not being observed statistical differences for spermatozoa cryopreserved on PCW-102c or TRIS. The morphometric parameters, however, suffered individual variations between the animals studied. The similarity of morphometric resulties of spermatozoa extended on PCW-102c and TRIS indicate that both have similar capacity of preserve the head structure of ram spermatozoa cryopreservated. It was recommended the research of new cryopreservation protocols that optimize the results obtained on the ram semen cryopreservation with the use of PCW-102c, the interaction of seminal constituents and spermatozoa membrane with the extenders components and the evaluation of ram semen cryopreserved on PCW-102c at artificial insemination programs.
