RESUMO

This trial was carried out from November 2005 to January 2006, in order to compare toxicity and the effect of the addition of cryoprotectant and activator solutions to curimba Prochilodus lineatus semen, before and after freezing. Semen samples of 10 breeders were analyzed and diluted in eight cryoprotectants solutions (1:4). These were composed for methanol or dimethyl sulphoxide (DMSO) in 5%, 7.5%, 10% and 12.5% (v:v) concentrations. Activators evaluated were distilled water and 1% Sodium bicarbonate (NaHCO3). A part of the diluted semen was frozen while the remaining fraction was maintained in rest and evaluated after 10 minutes. For freezing, leftover diluted semen was stored in 0.5 ml straws and placed in a dry shipper for 24 hours. Afterwards, they were immersed in liquid N2 where remained for 4 days. Milt samples were thawed by immersion in hot water bath (60ÂC) for 8 seconds. The DMSO estimated concentration of 7.84% and the activation for NaHCO3 1% were less toxicant to the curimba spermatozoids, propitiating the largest rates (P<0.01) and durations of their motility (P<0.05). The largest motility rate after thawing can be obtained (P<0.01) to 8.79% mean cryoprotectant concentration, while methanol (P<0.01) and DMSO (P<0.05), in the concentrations of 12.5% and 8.37%, respectively, can provide larger durations of the motility after freezing. Samples with DMSO incited the smallest (P<0.05) percentage of fractured and degenerate tail. There was a linear reduction (P<0.05) of 1.19% in the percentage of total pathologies with the increase in the cryoprotectant concentration. Larger (P<0.05) fertilization rates were obtained in the samples with methanol in relation to those with DMSO and the activator type did not influence it. Considering the variability of the fertilization analysis, 8.5% DMSO provides appropriate protection to the spermatic cells of curimba before and after freezing, being recommended its use in the routine of fish farming stations together semen activation for NaHCO3 1%.
