RESUMO

The purpose of this investigation was to evaluate how different contact methods of methylene blue dye solution 0, 5%, pH 7 for 24 hours, have influenced, in the reading of the demarcation leakage by tracer agent on the root dentin of human teeth. This study assesses 6 different methods: the dye agitation using the ultrasonic Gnatus, the agitation with the ultrasonic cleaner, the passive immersion, vacuum at 25 mm of mercury was applied for 10 minutes and then left immersed for 24 hours, vacuum at 30 mm of mercury and vacuum at 650 mm of mercury for 24 hours. The dye was applied to 60 one rooted teeth and left for 24 hours, the specimens were washed, dried and sectioned longitudinally and each root canal had been filled out with Lysanda paste. An image analyzer, type Q 550 IW of the mark Leica Qwin, was used in which a routine was set up for the acquisition of the images. In the sequence, a second routine was set up for working with the image acquired, detecting the different levels of blue of the dye at the sample. The image was edited by the form operator to demarcate the projections of the infiltration area in square millimeters. The results were analyzed statistically (ANOVA and Tukey). Considering the whole experiment and the different ways of impregnating the dye, none of the experimental conditions was better than the other. Comparing the different methods of dye impregnation among the different thirds separately, it resulted in a statistically significant difference in the results at the cervical thirds, where the vacuum at 30 mm of mercury was superior to the ultrasonic Gnatus and to the vacuum at 25 mm of mercury; In the medium thirds the passive immersion was superior in the results to the ultrasonic Gnatus. In relation to the apical third no experimental condition showed statistically significant difference. When the analysis was among the different thirds, the cervical third and medium third didn’t present difference, while, between the cervical and apical thirds and the medium and apical thirds there were statistically significant differences.
