RESUMO

Canatoxin (CNTX), a toxic protein extracted from the mature seeds of Canavalia ensiformis, induces secretion in several cells systems. Concanavalin A (Con A), the lectin that is also present in Canavalia ensiformis seeds, induces histamine secretion from rats mast cells. AIthough being two completely distinct principIes, CNTX and Con A share some pharmacological properties. The purpose of the present work was to verify if CNTX causes histamine release from rat peritoneaI mast cells and to perform a detaiIed study of this phenomenon. The ollowing resuIts were obtained: 1- CNTX induced histamine reIease from rat peritoneal mast cells; 2- The phenomenon was concentration (uM) and time-dependent; 3- The best conditions for the reaction to occur were at 37ºC and physiological pH; 4 - T.he reaction was inhibited in the absence of Ca++, Mg++ or glucose. Thus, we conclude that CNTX triggers histamine secretion from rat Mast cells via an enzymatic reaction, probably involving the activation of arachidonic acid metabolism.
