Estudo in vitro do efeito da propolis da Bahia (Mata Atlantica) sobre estreptococos do grupo mutans e na atividade das glucosiltransferases

RESUMO

Propolis is a natural beehive product, and its main constituents are polyphenolic compounds, such as flavonoids and cinnamic acid derivatives. However, propolis type 6, from Bahia (Atlantic forest, Northeastern, Brazil) presents an unusual chemical composition containing basically non-polar compounds; flavonoids were not detected in this sample. This study evaluated the effects of this propolis and its fractions on growth and adherence of mutans streptococci and on the activity of glucosyltransferases (GTFs). Ethanolic extract of propolis (EEP) and 4 fractions (hexane, chloroform, ethyl acetate, and ethanol) were obtained according to standard methods of chemical fractionation. Negative control was 80% ethanol (v/v). For the antibacterial assays, agar diffusion method, minimum inhibitory concentration (MIC), and minimal bactericidal concentration (MBC) against Streptococcus mutans Ingbritt 1600, S. sobrinus 6715 and two clinical isolates of each species were determined. Cell adherence of the microorganisms to a glass surface was measured spectrophotometrically at 550 nm. We also explored the effect of propolis and its fractions on the activity of purified GTFs in solution and adsorbed onto saliva-coated hydroxyapatite. The MIC values were 50-100 mu g/mL for S. mutans Ingbritt 1600, and 25-100 ?mu g/mL for S. sobrinus 6715. The cell adherence was also inhibited at concentrations as low as 25-50 mu g/mL Among the fractions, hexane (MIC 12.5-50 mu g/mL) and chloroform (MIC 25-100 mu g/mL) were the most active. MBC for EEPand fractions (hexane and chloroform) was 4-8 times higher than MIe. The activity of GTFs in solution was remarkably inhibited by EEP (88-96%), and by hexane and chloroform fractions (60-90% inhibition at 100 mu g/mL). In conclusion, the new variety of propolis showed remarkable inhibitory effects on bacterial growth, cell adherence and GTFs activity.
