RESUMO

The ability of snake venoms to induce local effects is well known. In this thesis, the oedema and hemorrhage induced in rats by heated and non-heated Bothrops lanceolatus venom (BL V) was investigated. Oedema was induced by the subplantar injection of venom (12.5-100 µg/paw) and was quantified by hydroplethysmography at various times thereafter. Hemorrhage was induced by injecting venom (0.25 - 64 µg) into the left gastrocnemius muscle and was expressed as accumulation of extravascular hemoglobin(hb) as determined by the cyanomethemoglobin method. With ali of the venom doses tested, the oedematogenic response peaked within 15 mino The maximal response was observed at a venom dose of 75 µg/paw (saline control = 0.09 ± 0.001 ml and venom = 1.09 ± 0.05 mi, n=7) and disappeared within 24h. The maximum hemorrhagic effect was observed with 32 µg of BLV (saline control = 1.23 ± 0.06 mghb/g of tissal and venom = 6.77 ± 0.69 mghb/g of tissal, n=6) and disappeared within 72h. Heating BLV (97°C, 30 sec) reduced the oedema by 70% and completely abolished the hemorrhagic activity. The combined administration of heated BL V (75 µg) With iloprost (0.1 µg) potentiated the oedema two-fold. Pre-treating the rats with mepyramine (6 mg/kg), cyproheptadine (6 mg/kg), mepyramine and cyproheptadine (6 mg/kg each, 15 min before), dexamethasone (1 mg/kg 2 h before), BW A4C (10 mg/kg) and Hoe 140 (0.6 mg/kg each 30 min before), inhibited the oedema induced by heated BL V (75 µg) plus iloprost (0.1 µg) by 28%, 55%, 77%, 63%, 35% and 30%; respectively. The co-administration of heated BL V (75 µg) with captopril (300 µg) potentiated the oedema by 44%, even in the absence of iloprost. Indomethacin (10 mg/kg) had no effect on the oedematogenic response. Pre-incubating native (non-heated) BL V with antiBL Vantiserum completely neutralized the hemorrhagic effect but only partially. abolished (45%) the oedema. The intravenous administration of antiBL V antiserum immediately after the subplantarar intramuscular injection of venom neutralized 30% of the oedema and 38% of the hemorrhage. When antiBLV antiserum was incubated with heated BLV plus iloprost and injected into rat paws, the neutralization was discrete (abaut 30%) and was observed only with the lowest proportion (0.125 ml antiBLV/mg BLV) tested. When administered intravenously immediately after the injection of heated venom, there was no neutralization whatsoever. These results indicate that non-heated BL V induces oedema in the paw and hemorrhage in the muscle of rats while heated BLV induces oedema without hemorrhage. AntiBLV antiserum completely neutralized the hemorrhagic effect and only partially inhibited the oedema when it was preincubated with BtV. The antiserum was less effective in neutralizing both activities when administered intravenously. These observations indicate that bradykinin, histamine, serotonin and lipoxygenase products are involved in the oedema caused by Bothrops lanceolatus venom
