RESUMO
Hyaluronic acid (HA) is a polysaccharide composed of 2000-25000 disaccaridic units of glucuronic acid and N-acetylglucosamine ?-bonds formed by 1-3 and 1-4 ?-turn. Due its physicochemical properties such as lubricity, hydrophilic and biocompatible, HA has been widely applied in the medical field. Many Studies show that production of HA by fermentation microbial allows optimization through cells nutritional requirements knowledge and control conditions cultivation as pH and temperature. The optimum pH for growth and production HA is 7.0, while the pH was neutral ("stress") which induces metabolic changes with repercussions on the production of HA. The effects of pH are poorly reported in the literature, especially regarding acid treatment in the region. In this paper, in first stage, were studied the effects of treatment in the inoculum pH 4.5, 5.0 and 5.5 and with times of 15, 30 and 60 minutes. We used the organic acid (Acetic) and inorganic (hydrochloric acid). The fermentations were conducted without forced aeration in flasks with pH 7.0. The best production of the AH (0.74 g / L) was obtained with treatment using hydrochloric acid, compared to treatment with acetic acid (0.49 g / L), and the inoculum untreated acid (0.31 g / L), used as control. In the second stage was studied the effect of the acid stress in the inoculum using stirred tank reactor with forced aeration in the best conditions determined in the first stage. It was observed an increased production of AH (2.5 g / L) when were used the conditions: the acid shock inoculum, pH 7.0 and 2vvm aeration during fermentation. And the third stage of the work was carried out in a study using cardiolipin added in the culture medium (in vivo). It was observed that cardiolipin plays an important role when we want to achieve a high molecular weight for AH
